cRaf-1 inhibitors to prevent AD pathology

Recently, we found that the enzyme, cRaf-1 is dysregulated in Alzheimer's disease (AD) brains and its inhibition prevents A&beta-induced neuronal cell death, and restores memory in old AD mice. The goal of the research proposed is to expand and confirm our preliminary data showing the use of Sorafenib to prevent or cure AD. We hypothesize that inhibition of cRaf-1 can inhibit the production of the amyloid peptide and its neuroinflammatory effects on the brain. These experiments should determine whether Sorafenib, a drug with minimal side effects, can recover memory deficit in AD.
