Heat Shock Protein Inducers for the Treatment of Alzheimer's Disease
Alzheimer's disease (AD) is a devastating degenerative disease imparting tremendous suffering to patients and caregivers alike. AD is characterized by insoluble deposits of the protein beta-amyloid. It has been shown that the chaperone heat shock protein HSP70 can clear the neurotoxic beta-amyloid oligomers which would be of great benefit for the treatment of AD. Since HSP70 administered itself intravenously is not expected to cross the blood brain barrier which is required for the treatment for AD, we have initiated an innovative program to identify small molecules that get into the brain and induce the synthesis of HSP70 and four other HSPs, by the use of a platform technology approach with five HSPs in four cell lines. In this way we expect to identify disease modifying therapeutic relief for AD as well as other protein aggregation disorders.
